چکیده
Heteromeric amino acid transporters are composed of a heavy and a light subunit linked by a disulfide bridge. 4F2hc/xCT elicits sodium-independent exchange of anionic L-cysteine and L-glutamate (system x(c)(-)). Based on the accessibility of single cysteines to 3-(N-maleimidylpropionyl)biocytin, we propose a topological model for xCT of 12 transmembrane domains with the N and C termini located inside the cell. This location of N and C termini was confirmed by immunofluorescence. Studies of biotinylation and accessibility to sulfhydryl reagents revealed a re-entrant loop within intracellular loops 2 and 3. Residues His(110) and Thr(112), facing outside, are located at the apex of the re-entrant loop. Biotinylation of H110C was blocked by xCT substrates, by the nontransportable inhibitor (S)-4-carboxyphenylglycine, and by the impermeable reagent (2-sulfonatoethyl) methanethiosulfonate, which produced an inactivation of H110C that was protected by L-glutamate and L-cysteine with an IC(50) similar to the K(m). Protection was temperatureindependent. The data indicate that His(110) may lie close to the substrate binding/permeation pathway of xCT. The membrane topology of xCT could serve as a model for other light subunits of heteromeric amino acid transporters.

Explaining calcium-dependent gating of anoctamin-1 chloride channels requires a revised topology.
RATIONALE Ca2+ -activated Cl channels play pivotal roles in the cardiovascular system. They regulate vascular smooth muscle tone and participate in cardiac action potential repolarization in some species. Ca2+ -activated Cl channels were recently discovered to be encoded by members of the anoctamin (Ano, also called Tmem16) superfamily, but the mechanisms of Ano1 gating by Ca2+ remain enigmatic...
متن کاملA Scheduling Model for the Re-entrant Manufacturing System and Its Optimization by NSGA-II
In this study, a two-objective mixed-integer linear programming model (MILP) for multi-product re-entrant flow shop scheduling problem has been designed. As a result, two objectives are considered. One of them is maximization of the production rate and the other is the minimization of processing time. The system has m stations and can process several products in a moment. The re-entrant flow sho...
متن کاملSolving Re-entrant No-wait Flexible Flowshop Scheduling Problem; Using the Bottleneck-based Heuristic and Genetic Algorithm
In this paper, we study the re-entrant no-wait flexible flowshop scheduling problem with makespan minimization objective and then consider two parallel machines for each stage. The main characteristic of a re-entrant environment is that at least one job is likely to visit certain stages more than once during the process. The no-wait property describes a situation in which every job has its own ...
متن کاملGlutamate receptor pores.
Glutamate receptors are ligand-gated ion channels that mediate fast excitatory synaptic transmission throughout the central nervous system. Functional receptors are homo- or heteromeric tetramers with each subunit contributing a re-entrant pore loop that dips into the membrane from the cytoplasmic side. The pore loops form a narrow constriction near their apex with a wide vestibule toward the c...
متن کاملCOMPREHENSIVE INVITED REVIEW The Cystine/Glutamate Antiporter System xc - in Health and Disease: From Molecular Mechanisms to Novel Therapeutic Opportunities
The antiporter system xc imports the amino acid cystine, the oxidized form of cysteine, into cells with a 1:1 counter-transport of glutamate. It is composed of a light chain, xCT, and a heavy chain, 4F2 heavy chain (4F2hc), and, thus, belongs to the family of heterodimeric amino acid transporters. Cysteine is the rate-limiting substrate for the important antioxidant glutathione (GSH) and, along...
